      目的:探究解郁合欢汤(Hehuan decoction for relieving dePression)在慢性应激引起的抑郁症模型小鼠中的抗抑郁作用及其潜在机制。 方法:采用正常小鼠和抑郁模型小鼠(慢性不可预见性应激造模,CUMS),研究解郁合欢汤不同剂量组(4,8,16g/kg)和阳性药氟西汀组对小鼠脑比重、行为学、五羟色胺(5-hydroxytryPtamine,5-HT)、去甲肾上腺素(NorePinePhrine,NE)、多巴胺(doPamine ,DA)和组织形态影响。 结果:与正常组比较,模型组脑比重、糖水偏好率、敞箱活动次数均下降(P<0.01),悬尾、强迫游泳不动时间延长,5-HT、DA、NE等单胺神经递质含量下降(P<0.01)。解郁合欢汤高、中、低剂量组脑比重、糖水偏好与模型组比较明显升高(P<0.05,P<0.01),解郁合欢汤高、中、低剂量组悬尾不动时间、强迫游泳不动时间与模型组比较明显降低(P<0.05,P<0.01)。敞箱实验中,解郁合欢汤高、中剂量组水平运动和垂直运动次数与模型组比较升高(P<0.05,P<0.01),但低剂量组与模型组比较无统计学差异(P>0.05)。解郁合欢汤高剂量组、中剂量组、氟西汀组脑5-HT、DA、NE,低剂量组的NE显著高于模型组,其中低剂量组脑5-HT、DA与模型组比较无统计学差异(P>0.05)。 结论:解郁合欢汤可能通过调节单胺类神经递质发挥抗抑郁作用,显示出潜在的治疗抑郁症的益处,可以推测:解郁合欢汤对于改善抑郁症状还有其他机制的协同作用,但尚未完全明确,有待进一步深入研究。

      Objective: To investigate the antidepressant effect of Hehuan decoction for relieving dePression in chronic stress-induced depression model mice and its potential mechanism. METHODS:Normal mice and depression model mice (Chronic Unpredictable Stress Modeling, CUMS) were used to study the effects of Hehuan decoction for relieving dePression on specific gravity of the brain, behavior, 5-hydroxytryPtamine (5-HT), norepinephrine, NE, dopamine, dopamine, and norepinephrine in the mice at different dosages of Hehuan decoction for relieving dePression and the positive dose of fluoxetine in mice. NE), dopamine (doPamine ,DA) and tissue morphology effects. RESULTS: Compared with the normal group, brain specific gravity, sugar-water preference rate, and the number of open-box activities were decreased in the model group (P<0.01), and the time of tail suspension and forced swimming immobilization was prolonged, and the content of monoamine neurotransmitters, such as 5-HT, DA, and NE, was decreased (P<0.01). Brain specific gravity and sugar-water preference in the high, medium and low dose groups of Undulant and Acacia Tang were significantly higher compared with the model group (P < 0.05, P < 0.01), and hanging tail immobilization time and forced swimming immobilization time in the high, medium and low dose groups of Undulant and Acacia Tang were significantly lower compared with the model group (P < 0.05, P < 0.01). In the open-box experiment, the number of horizontal and vertical movements was elevated in the high and middle dosage groups of Xieyu Hecheng Tang compared with the model group (P < 0.05, P < 0.01), but there was no statistical difference in the low dosage group compared with the model group (P > 0.05). Brain 5-HT, DA, and NE in the high-dose group, medium-dose group, and fluoxetine group of Xieyu Hecheng Tang, and NE in the low-dose group were significantly higher than those in the model group, in which there was no statistically significant difference between the brain 5-HT and DA in the low-dose group compared with the model group (P > 0.05). CONCLUSION: The antidepressant effect of Xieyu Heping Tang may be exerted through the regulation of monoamine neurotransmitters, which shows the potential therapeutic benefit for depression, it can be speculated that: Xieyu Heping Tang also has a synergistic effect of other mechanisms on the improvement of the symptoms of depression, but it has not yet been fully clarified, and needs to be further investigated in-depth.
