| 目的 探讨标本配穴针刺对胚胎着床障碍大鼠TH1/TH2平衡表达的影响。方法 将40只早孕大鼠随机分为正常组(Control,C)、模型组(Model,M)、黄体酮组(Progesterone,P)和标本配穴针刺组(Acupunture,A),每组各10只。M组、P组和A组给予米非司酮麻油溶液皮下注射造模。自造模当天起,A组给予标本配穴针刺治疗,P组给予黄体酮麻油溶液皮下注射治疗。造模干预后第8d颈椎脱臼处死各组大鼠,观察各组大鼠胚胎着床数并采用苏木精-伊红(H-E)染色法观察各组大鼠子宫内膜的组织形态学变化,采用酶联免疫吸附试验(ELISA)检测各组大鼠血清IL-10、IL-4、INF-γ、TNF-α的含量,采用蛋白印迹法(Western blot)检测各组大鼠子宫内膜IL-10、IL-4、INF-γ、TNF-α的蛋白表达,采用实时荧光定量聚合酶链式反应(Rt-PCR)检测各组大鼠子宫内膜IL-10、IL-4、INF-γ、TNF-α的mRNA表达。结果 各组在上述指标的比较均具有显著差异(P<0.05),两两比较结果显示,与C组相比,M组、A组及P组大鼠胚胎着床数均显著减少 (P<0.05),血清IL-4、IL-10的含量均显著降低(P<0.05),子宫内膜IL-4、IL-10的蛋白与mRNA表达均显著降低 (P<0.05),而血清INF-γ、TNF-α的含量均显著升高 (P<0.05),子宫内膜INF-γ、TNF-α的蛋白与mRNA均显著升高 (P<0.05);与M组相比,A组与P组的胚胎着床数均明显增加 (P<0.05),子宫内膜形态改善,血清IL-4、IL-10的含量均显著升高(P<0.05),子宫内膜IL-4、IL-10的蛋白与mRNA表达均显著升高 (P<0.05),而血清INF-γ、TNF-α的含量均显著降低(P<0.05),子宫内膜INF-γ、TNF-α的蛋白与mRNA均显著降低(P<0.05);与P组相比,A组的胚胎着床数、子宫内膜形态、血清IL-10、IL-4、INF-γ、TNF-α的含量及子宫内膜IL-10、IL-4、INF-γ、TNF-α的蛋白与mRNA表达均无显著差异(P>0.05)。 |

| Objective To explore the effect of “Biao-Ben acupoint combination”acupuncture on the expression of TH1/TH2 in rats with embryo implantation failure. Methods 40 early pregnant rats were randomly divided into Control group (C), Model group (M), Progesterone group (P) and acupuncture group with “Biao-Ben acupoint combination” Acupuncture(A), with 10 rats in each group. The model was made by subcutaneous injection of mifepristone sesame oil solution in group M, group P and group A. From the day of modeling, group A was given acupuncture treatment with “Biao-Ben acupoint combination” and group P was treated with subcutaneous injection of progesterone sesame oil solution. The rats in each group were killed on the 8th day after cervical vertebra dislocation.The number of embryo implantation in each group was observed and the contents of IL-10, IL-4, INF-γ and TNF-α in serum were detected by enzyme linked immunosorbent assay (ELISA).The protein expressions of IL-10, IL-4, INF-γ and TNF-α in endometrium of rats in each group were detected by Western blot. Real-time fluorescence quantitative polymerase chain reaction (Rt-PCR) was used to detect the mRNA expression of IL-10, IL-4, INF-γ and TNF-α in endometrium of rats in each group.Results There were significant differences in the above indexes among all groups(P<0.05).The results showed that compared with group C, the number of embryo implantation in group M, group A and group P decreased significantly(P<0.05), the contents of IL-4 and IL-10 in serum decreased significantly(P<0.05), and the protein and mRNA expression of IL-4, IL-10 and mRNA in endometrium decreased significantly(P<0.05).However, the contents of INF-γ and TNF-α in serum, the protein and mRNA of INF-γ and TNF-α in endometrium were significantly increased (P<0.05). Compared with group M, the number of embryo implantation in group A and group P increased significantly(P<0.05), the contents of IL-4 and IL-10 in serum increased significantly(P<0.05), the expression of IL-4, IL-10 and mRNA in endometrium increased significantly(P<0.05), while the contents of INF-γ and TNF-α in serum decreased significantly(P<0.05), and the protein and mRNA of INF- γ and TNF- α in endometrium decreased significantly(P<0.05). Compared with group P, there was no significant difference in the number of embryo implantation, the content of serum IL-10, IL-4, INF-γ, TNF-α and the expression of IL-10, IL-4, INF-γ, TNF-α protein and mRNA in endometrium in group A(P>0.05). Conclusion “Biao-Ben acupoint combination”acupuncture can significantly increase the number of embryo implantation in rats with embryo implantation failure, and its mechanism may be related to the up-regulation of IL-4, IL-10 and down-regulation of the expression of INF-γ and TNF-α in serum and endometrium, and the restoration of the balance between the expression of Th1 and Th2 cytokines. |
