      目的:通过临床观察和网络药理学方法探析癃清胶囊抗尿路感染疗效及作用机制。方法:选取西医确诊为单纯性尿路感染,且中医辨证为下焦湿热证的病人60例,随机分为对照组和试验组,每组30例。试验组口服癃清胶囊,对照组口服癃清胶囊模拟剂。治疗7天后,考察两组病人中医证候积分、生活质量评分、单项症状消失时间及安全性。同时,利用Swiss Target Prediction和TCMSP数据库获取癃清胶囊的活性成分及潜在作用靶点,通过GeneCards数据库获取尿路感的相关靶点,筛选共同靶点,运用Cytoscape构建“活性成分-靶点”网络;通过STRlNG数据库构建PPI网络并进行KEGG和GO分析。结果:与对照组病人相比,试验组病人中医证候积分、生活质量评分、单项症状消失时间较均有所改善(P<0.05)。网络药理学结果显示癃清胶囊作用于尿路感染的生物学过程主要在蛋白磷酸化、凋亡和增殖过程以及对外刺激的应答等方面;主要富集在癌相关通路、T细胞受体信号通路等信号通路上。结论:癃清胶囊可能通过癌相关通路等信号通路发挥抗尿路感染作用,可改善病人临床症状,降低中医证候积分,改善尿频、尿急、尿痛等症状。

      Objective Antiurinary tract infection efficacy and mechanism of action of Longqing Capsule (LQC) by clinical observation and network pharmacological methods. Methods Sixty patients who were diagnosed as simple urinary tract infection by western medicine and identified as Lower Jiao Damp-Heat syndrome by traditional Chinese medicine were selected and randomly divided into the control group and the experimental group, with 30 cases in each group. The test group took LQC orally and the control group took LQC analogue orally. After 7 days of treatment, the TCM evidence points, quality of life scores, time to disappearance of single symptom and safety were examined in both groups. At the same time, the Swiss Target Prediction server and TCMSP database were used to obtain the active ingredients and potential targets of LQC, and the GeneCards database was used to obtain the targets related to urolithiasis, and the common targets were screened out, so as to construct the network of active ingredients-targets by using Cytoscape. Construction of PPI networks and KEGG and GO analyses via the STRlNG database. Results Compared with the patients in the control group, the patients in the experimental group showed improvement in TCM evidence points, quality of life scores, and time to disappearance of single symptom than all (P < 0.05). The results of network pharmacology showed that the biological process of LQC in urinary tract infection is mainly in protein phosphorylation, apoptosis and proliferation process, and the response to external stimuli; mainly enriched in the cancer-related pathway, T-cell receptor signalling pathway and other signalling pathways. Conclusion LQC may play an anti-urinary tract infection effect through the cancer-related pathway and other signalling pathways, which may improve the clinical symptoms of patients, reduce the Chinese medicine evidence points, and improve the symptoms of urinary frequency, urgency, and urinary pain.
