      目的:基于网络药理学探究消瘰丸治疗甲状腺结节的潜在作用机制。方法:运用多个数据库检索并筛选出消瘰丸活性成分,将筛选出的消瘰丸活性成分导入Swiss Target Prediction数据库筛选出药物靶点。在Gene Cards、Omim和Therapeutic Target Database数据库上检索并筛选出甲状腺结节疾病靶点,运用Venny将药物靶点和疾病靶点进行交集,得到消瘰丸治疗甲状腺结节的潜在作用靶点。将药物与疾病的交集靶点导入String数据库进行蛋白质-蛋白质相互作用(PPI)分析,并运用Sytoscape软件进行可视化及拓扑分析,筛选出核心靶点。并使用DAVID数据库对交集靶点进行GO与KEGG富集分析。 结果:共筛选出药物靶点359个,疾病靶点3652个,交集靶点185个。PPI网络分析得到消瘰丸治疗甲状腺结节的核心靶点有AKT1、TP53、BCL-2、SRC、EGFR等。GO富集分析结果显示,潜在治疗靶点主要参与了蛋白质磷酸化、凋亡的负调控、对异源性刺激的反应和信号传导等生物过程;KEGG富集分析结果显示消瘰丸治疗甲状腺结节涉及到的通路主要包括癌症通路、甲状腺激素通路、PI3K-Akt通路、化学致癌-受体激活通路等。 结论:消瘰丸的多种活性成分能够通过多靶点、多通路协同作用,对甲状腺结节起到治疗作用,其机制可能涉及到癌症通路、甲状腺激素通路、PI3K-Akt通路、化学致癌-受体激活通路等信号通路的调控。

      Objective: To explore the potential mechanism of Xiaoluo pills in the treatment of thyroid nodules based on network pharmacology.? Methods: We searched and screened the active ingredients of Xiaoluo pills through multiple databases, and then imported them into the Swiss Target Prediction database to screen out the drug targets. The targets of thyroid nodules were retrieved and screened on the Gene Cards, Omim and Therapeutic Target Databases, and we used Venny to intersect drug targets and disease targets to obtain potential targets for the treatment of thyroid nodules. The intersection targets of drugs and diseases were imported into the String database for protein-protein interaction (PPI) analysis, and Sytoscape was used for visualization and topology analysis to screen out the core targets. And we used the David database for GO and KEGG enrichment analysis of intersecting targets.? Results: A total of 359 drug targets, 3652 disease targets, and 185 intersecting targets were screened. The core targets of Xiaoluo pills in the treatment of thyroid nodules were AKT1, TP53, BCL-2, SRC, EGFR and so on. The results of GO enrichment analysis showed that the potential therapeutic targets were mainly involved in biological processes such as protein phosphorylation, negative regulation of apoptotic process, response to xenobiotic stimulus and signal transduction, and KEGG enrichment analysis showed that the pathways involved in the treatment of thyroid nodules mainly included cancer pathway, thyroid hormone signaling pathway, PI3K-Akt pathway, and chemocarcinogenic-receptor activation pathway.? Conclusion: A variety of active ingredients of Xiaoluo pills can play a therapeutic role in the treatment of thyroid nodules through multi-target and multi-pathway synergistic effects, and its mechanism may may involve the regulation of cancer pathway, thyroid hormone signaling pathway,PI3K-Akt pathway, and chemical carcinogenic-receptor activation pathway.
